There are many types of phosphate buffers available. However, the most commonly used phosphate buffer consists of a mixture of monobasic dihydrogen phosphate and dibasic Mono hydrogen phosphate.

 

By varying the amount of each salt, a range of buffers can be prepared that buffer well between pH 5.8 and pH 8.0. Phosphates have a very high buffering capacity and are highly soluble in water.

 

Process of Preparation of 0.067M Phosphate Buffer:

  • Dissolve 3.532 gm of potassium dihydrogen phosphate and 14.542 gm of disodium hydrogen phosphate in water.
  • Add sufficient water to make the solution to 1000 ml.

 

*The phosphate buffer solution bottle should be open for the minimum time required to dispense the solution.

 

After use, the bottle should be tightly recapped and stored under normal laboratory conditions.
